Study on the efficiencies of the mobilization, collection and the safety of the transplantation of autologous peripheral blood stem cell for myocardial infarction patients

Abstract ] Objective Toinvestigatetheefficienciesof mobilization andcollection of
autologousperipheralbloodstemcell ( APBSC ) formyocardialinfarctionpatients.Andtostudy
thesafetyandfeasibilityofmyocardialinfarctiontreatmentviatheautologousperipheralblood
stemcelltransplantation ( APBSCT ) .Methods Forty-sixmyocardialinfarctionpatientswiththe herapyofconventionaldrugweregivenrecombinanthumangranulocytecolonystimulatingfactor
(
rhG-CSF ) 450 μ g / d ( ≤65kg ) or600 μ g / d ( >65kg ) bysubcutaneousinjectionatfivedays
beforecoronaryarterybypassgraft ( CABG ), accordingtopatient'sbodymass , thenwerecarried
onthe APBSC mobilizationandexpansion.APBSCs werecollected24hbeforeCABG.All
patientsunderwentCABGwithnon-cardiopulmonaryby-pass , andtheAPBSCswereinjectedvia
thebridgecoronary vesselsandsub-epicardially.Beforeandafter APBSC mobilizationand
collection , mononuclearcell ( MNC ) andCD34
+
cellfromtheperipheralbloodweretested , and
thebloodpressure , heartrate , respiratory , whitebloodcell ( WBC ), redbloodcell ( RBC ),
hemoglobin ( Hb ), hematocrit ( Hct ), platelet ( Plt ) andserum Ca

werealsotested.Adverse
reactionsintheprocessofAPBSC mobilization , collectionandtransplantation wereobserved.
Results ThenumbersofWBC , MNCandCD34
+
cellsinperipheralbloodwereallsignificantly
increased ( P <0.01 ) after APBSC mobilizationthanthatbefore APBSC mobilization.After
APBSCcollection , thenumbersofMNCandCD34
+ cellsweresignificantlyincreased (
P <0.01 ) .
Nosignificanceexistedinthebloodpressure , heartrate , respiratory , WBC , RBC , Hb , Hctand
serum Ca
2+
( P >0.05 ) .ThenumberofPlt wassignificantly ( P <0.01 ) decreasedafter
collection.Adversereactionsoccurredin20casesduring APBSC mobilization , collectionand
transplantation , includingtheadversereactionof17.4% ( 8 / 46 ) inAPBSCmobilization , 10.9%
(
5 / 46 ) inAPBSCcollection , and15.2% ( 7 / 46 ) intransplantation.Conclusion TheMNCand
CD34
+
cellscanbegreatlyincreasedafterAPBSC mobilizationandcollection , indicatingsafty
andfeasibilityofAPBSCTformyocardialinfarction.
