Detection and clinical significance of methylation in the promoter region of SFRP1 and SFRP2 genes in renal clear cell carcinoma#br#

Abstract: [Abstract]〓Objective〖HTSS〗〓To investigate the promoter methylation status of secreted frizzledrelated protein 1 (SFRP1) and SFRP2 gene in renal clear cell carcinoma(RCCC).
〖HTH〗〖WTHZ〗Methods〖HTSS〗〓MSP was used to examine the methylation status of SFRP1 and SFRP2 gene in RCCC tissues and corresponding adjacent noncancerous tissues. The expression of βcatenin in Wnt pathway was detected by immunohistochemical streptavidin/peroxidase(SP) method.
〖HTH〗〖WTHZ〗Results〖HTSS〗〓Methylation frequencies of SFRP1 and  SFRP2 gene in 122 cases of RCCC were significantly higher than that in corresponding adjacent noncancerous tissues(P<005). The incidence of methylation of SFRP1 gene in stage Ⅳ was significantly higher than that in stage Ⅰ, Ⅱ and Ⅲ in RCCC tissues(P<005). The expression of βcatenin in RCCC tissues was significantly higher than that in corresponding adjacent noncancerous tissues(P<005). The expression of βcatenin in hypermethylated SFRP1 and SFRP2 gene in RCCC tissues was significantly higher than that in unmethylated RCCC tissues(P<005).
〖HTH〗〖WTHZ〗Conclusion〖HTSS〗〓SFRP1, SFRP2 gene methylation may be involved in the occurrence of RCCC mediated by Wnt/βcatenin signal transduction pathway, and high methylation of SFRP1 gene was associated with invasion and metastasis of RCCC.
