Correlation between COX-2 and survivin expression in benign and malignant pleomorphic adenomas of parotid gland

Abstract: [Abstract]Objective〖HTSS〗 To investigate the relationship between COX-2 and survivin expression in the development and progression of malignant adenoma of the parotid gland, through detecting the expression of COX-2 and survivin protein in pleomorphic adenoma and malignant pleomorphic adenoma.
  〖WTHZ〗Methods〖HTSS〗According to the Seifert classification criteria, the pleomorphic adenoma specimens were divided into four groups: group A, 20 samples categorized as myxoid type nodules(predominance of intervening stroma); group B, 24 samples categorized as other type; group C, 22 samples categorized as cell-rich type nodules(predominance of epithelial cells); group D, 18 samples categorized as malignant pleomorphic adenoma. The expression of COX-2 and survivin in each group was detected by immunohistochemistry method, and the correlation between the expression of COX-2 and survivin in the occurrence and prognosis of parotid pleomorphic adenoma was analyzed.
  〖WTHZ〗Results〖HTSS〗COX-2 protein was expressed in the cytoplasm of tumor cells and occasionally in the cell membrane and nucleus. The positive expression rate of COX-2 protein in group C and group D was higher than that in group A(P<0.05). Survivin protein was expressed in glandular duct epithelial cells and mucoid tissues. Brown granules were found in cytoplasm or nucleus. The positive expression rate of Survivin in group D was higher than that in group A(P<0.05). Through Spearman correlation analysis, COX-2 expression was positively correlated with survivin expression(rs=0.560, P<0.01). The higher the expression level of survivin, the higher the expression level of COX-2.
  〖WTHZ〗Conclusion〖HTSS〗The expression of COX-2 and survivin in benign and malignant pleomorphic adenomas of the parotid gland is correlated, It provides a theoretical basis for the clinical pathological diagnosis of pleomorphic adenoma, and may become a new biomarker and therapeutic gene target.
