Abstract ] Objective ToexplortheeffectsofD-galactoseonthecognitivequalitydecline
andtheneuronalplasticityintheprocessofagingbyobservingthechangesoflearningand
memoryqualityandtheexpressionofgamma-aminobutyricacidreceptor1 ( GABAR1 ) andN-
methyl-D-aspartatereceptor2B ( NMDAR2B ) inrats.Methods MaleSDratswererandomly
dividedintoexperimentalgroup ( 10 ) andcontrolgroup ( 20 ) .D-galactose ( 15mg ·
kg
-1 · d -1 )
wassubcutaneouslyinjected for 10 weeksin experimental group and the same dose of
physiologicalsalinewasgiventocontrolgroup.Morriswater mazetestwasperformedafter
modelprepared.ThenthebraintissuewascollectedandthemRNAtranscriptionandprotein
expression of GABAR1 and NMDAR2B were detected by in situ hybridization and
immunohistochemistrymethods.Results Thelatencywassignificantlylongerandthefrequencyofpassingthroughthetargetwassignificantlylessinexperimentalgroupthanthoseinthe
controlgroup ( P <0.05 ) .GABAR1andNMDAR2BwerepositivelyexpressedatthemRNAand
proteinlevelineachgroup.Theproteinexpressionand mRNAtranscriptionofNMDAR2Bin
experimentalgroupwereweakerthanthoseinthecontrolgroup.Buttheproteinexpressionand
mRNAtranscriptionofGABAR1werehigherinexperimentgroupthanincontrolgroup , the
differencewasstatisticallysignificant ( P <0.05 ) .Conclusion Thelearningandmemoryquality
declinewererelatedtotheexpressionchangeofneurotransmitterreceptorssuchasNMDAR2B
andGABAR1inhippocampusinthebrainagingprocessofSDrats.
