Effect of androgen on telomeres length in aplastic anemia patients#br#

Abstract: [Abstract]Objective〖HTSS〗To explore the effect of androgen on telomeres in aplastic anemia(AA) patients, and the effect of telomeres in the pathogenesis of AA.
Methods〖HTSS〗The patients were collected according to the AA clinical diagnostic criteria. They were divided into two groups, cyclosporin A(CsA)+androgen and CsA respectively, 15 cases in each group. The genomic DNA of peripheral blood was extracted by nucleic acid extractor and nucleic acid extraction kit. The relative length of telomere in peripheral blood leukocytes was measured by real-time fluorescence quantitative PCR and compared with that of 15 healthy controls.
Results〖HTSS〗The relative T/S ratio of the telomere length in AA patients was significantly shorter than that in the normal control group(P<0.05). After treatment, the relative T/S ratio of the telomere length in the CsA group was reduced volatility,and that of the CsA+androgen group was increased. The relative T/S ratio of the telomere length in the CsA+androgen group was evidently higher than that in the CsA group.The differences between groups,time points, interaction of groups and time points were statistically significant(P<0.05). The hematological improvement of CsA+androgen was better than that of CsA group at 12 months after treatment(P<0.05).
Conclusion〖HTSS〗Androgen can regulate telomere length in AA patients, and the length of telomeres increased after androgen therapy. Androgen is effective in the treatment of AA with telomere dysfunction, and clinical hematologic improvement can be achieved.
