Correlation study between serumal expression of miRNA-21 and miRNA-192, and diabetic nephropathy#br#

Abstract: [Abstract]Objective〖HTSS〗To investicate the correlation between serumal expression of miRNA-21 and miRNA-192,and diabetic nephropathy(DN).
  〖WTHZ〗Methods〖HTSS〗These patients with type 2 diabetes mellitus(38 cases, DM group) and DN(27 cases,DN group),as well as 30 healthy check-up population were performed detecting their serumal expression of miRNA-21 and miRNA-192 by real time-PCR(RT-PCR). Blood flow-related parameters such as peak velocity(PSV) of renal aorta(MRA) and resistance index(RI) were measured by PHILIPS-IU22 color Doppler ultrasound system.The relationship between the above indexes and DN would be described via the comparative analysis.
  〖WTHZ〗Results〖HTSS〗RI of DN group and DM group was significantly higher than that of control group, and RI of DN group was significantly higher than that of DM group, PSV of DN group was significantly lower than that of DM group and control group(P<0.05). The expression of miRNA-21 increased whereas expression of miRNA-192 decreased in DN group and DM group compared with those in control group, and a same trend towards expression of miRNA-21 and miRNA-192 was also found in DN group compared with those in DM group(P<0.05). In DN group, the expression of miRNA-21 and miRNA-192 was negatively correlated(r=-0.837, P<0.05). The expression of miRNA-21 was positively correlated with the levels of RI(r=0.812, P<0.05), and was negatively correlated with the level of PSV(r= -0.793, P<0.05) . The expression of miRNA-192 was negatively correlated with the levels of RI(r=-0.807, P<0.05), and was positively correlated with the level of PSV(r=0.782, P<0.05). In DM group, the expression of miRNA-21 and miRNA-192 was negatively correlated(r=-0.816, P<0.05); miRNA-21 was positively correlated with RI level(r=0.714, P<0.05); miRNA-192 was negatively correlated with RI level(r=-0.745, P<0.05).
  〖WTHZ〗Conclusion〖HTSS〗The expression of miRNA-21 and miRNA-192 in serum is related to the occurrence and development of DN. Detecting the expression of miRNA-21 and miRNA-192 can provide the gene basis for early identification and treatment of DN.
