补肾活血固齿方对MC3T3E1细胞BMP2表达及TRPV5/TRPV6通道影响的实验研究

摘要: [摘要] 目的 观察补肾活血固齿方对小鼠颅顶前成骨细胞( MC3T3-E1 细胞)骨形态发生蛋白 2 ( bone
morphogeneticprotein2 , BMP2 )表达及 TRPV5 / TRPV6 通道的影响,探讨补肾活血固齿方成骨的作用机制。方法
按体表面积折算等效剂量的补肾活血固齿方灌胃大鼠,制备含药血清;等剂量生理盐水灌胃大鼠,制备无药血清作
为对照。用 10% 含药血清、无药血清分别培养 MC3T3-E1 细胞 24 、 48 、 72h , RT-PCR 法检测 MC3T3-E1 细胞
BMP2 、 TRPV5 / TRPV6mRNA 的表达。结果 含药血清组 MC3T3-E1 细胞的 BMP2 、 TRPV6mRNA 表达量较无
药血清组增加( P <0.05 ),且随时间延长有逐渐增加趋势,但差异无统计学意义(
P >0.05 )。含药血清组和无药血
清组 MC3T3-E1 细胞 中 均未 检测 到 TRPV5 mRN

Abstract:
[Abstract ] Objective ToobservetheeffectofBushen HuoxueGuchiFormulaonbone
morphogeneticprotein2 ( BMP2 ) andTRPV5 / TRPV6channelexpressions , andtoexplorethe
mechanism ofBushen Huoxue GuchiFormulainboneformation.Methods Herb-contained
serum waspreparedfromratsbygavagewithBushen HuoxueGuchiFormulabyusingbody
surfaceareaconvertedequivalentdose.Blank-containedserumascontrolgroupwaspreparedby
gavagewithnormalsaline.TheBMP2andTRPV5 / TRPV6mRNAexpressionsweredetectedin
MC3T3-E1cellsby RT-PCRafterthecellsweretreated with10% herb-containedserum or
blank-containedserumfor24h , 48hand72h , respectively.Results BMP2 , TRPV6mRNA
weresignificantlyincreasedin herb-containedserum treatmentgroupcompared with blank-containedserum treatmentgroup ( P <0.05 ) .Withtheprolongation oftime , the mRNA
expressionof BMP2 and TRPV6 had a graguallyincreased tendency , butthere was no
statisticallysignificantdifference ( P >0.05 ) .TRPV5 mRNA wasnotdetectedeitherinherb-
containedserumtreatmentgrouporblank-containedserumtreatmentgroup.Conclusion Bushen
HuoxueGuchiFormulamayupregulateBMP2expressionthroughTRPV6channelstoinduce
differentiationofosteoblast.
