丁苯酞联合依达拉奉对局灶性脑梗死大鼠皮质血脑屏障及咬合蛋白表达的影响

摘要: [摘要] 目的 探讨丁苯酞联合依达拉奉对局灶性脑梗死大鼠皮质血脑屏障及咬合蛋白表达的影响。方法
选取成年 SD 雄性大鼠 60 只,随机分为对照组、模型组、丁苯酞组、丁苯酞联合依达拉奉组,每组 15 只。各组于模
型制作成功后 72h 采用伊文思兰渗出法检测大鼠血脑屏障通透性变化,免疫组织化学和 Westernblot 检测损伤区
周围皮质咬合蛋白表达变化。结果 与对照组比较,模型组、丁苯酞组、丁苯酞联合依达拉奉组梗死区大脑皮层血
脑屏障通透性均增加,皮质咬合蛋白表达均减少。与模型组相比,丁苯酞组和丁苯酞联合依达拉奉组皮质脑组织
伊文思蓝渗出量减少,皮质咬合蛋白表达增加。与丁苯酞组比较,丁苯酞联合依达拉奉组伊文思蓝渗出量减少,咬
合蛋白表达量增加。结论 丁苯酞软胶囊可保护血脑屏障,增加大脑皮质咬合蛋白的表达。与依达拉奉联合应用
保护血脑屏障、增加咬合蛋白表达作用更明显。

Abstract: [
Abstract ] Objective Toexploretheeffectsofbutylphthalidecombinedwithedaravoneon
blood-brainbarrierpermeabilityandoccludinexpressionsincortexoffocalcerebralinfarctionin
rats.Methods Sixtysprague-dawleymaleratswererandomlydividedintofourgroups : control
group , modelgroup , butylphthalidegroupandbutylphthalidecombinedwithedaravonegroup ,
with15ratsineachgroup.Themodelwasestablishedbasedontheprincipleofphotochemical
initiationofthrombosis.At72hafterinjury , evansblueextravsationmethodwasusedtodetect
theblood-brain barrierpermeability.Immunohistochemistryand western blot wereusedto
examinethecortexoccludinexpressionaroundtheinfarctionarea.Results Comparedwiththe
controlgroup , theblood-brainbarrierpermeabilitiesinthemodelgroup
, butylphthalidegroup
andthebutylphthalidecombined withedaravonegroup wereincreasedsignificantly.Andthe
occludinexpressionsaroudtheinfarction wereupregulatedsignificantly.Compared withthe
modelgroup : evans blueextravsationsin cortexin the butylphthalide group andin the butylphthalidecombined withedaravonegroup weredecreased , andtheoccludinexpressions
aroudtheinfarctionareawereincreased.Comparedwiththethebutylphthalidegroup : evansblue
extravsationincortexinthebutylphthalidecombinedwithedaravonegroupwasdecreased , and
theoccludinexpressionaroudtheinfarction wasincreased.Conclusion Butylphthalidecould
protectthebolld-brainbarrier , andincreasetheexpressionofoccludin.Theeffectwas more
obviousbyusingthecombinationofbutylphthalideandedaravone.
