脐血造血细胞体外扩增及其粘附特性变化

摘要: 目的 探讨脐血造血细胞体外扩增过程中的粘附特性变化,为扩增最佳时机的选择提供实验依据。方法 用长期培养法体外扩增脐血造血细胞,并用流式细胞仪测定其表面粘附分子变化。结果  CD11b表达于第10天显著增加,第30天达34.3%;CD44表达于第20天达最高水平;CD54表达则于第5天明显增加,第30天达14.9%;CD34+造血细胞在第15天达高峰,之后渐下降,并维持在一定水平。CD44及CD54表达与CD34+造血细胞变化存在相关性。结论 脐血造血细胞体外扩增过程中,CD44和CD54的持续高表达有利于造血细胞移植后的归巢;移植的最佳时机以长期培养第14天为宜。

Abstract: Objective To investigate the variation of cell adhesion molecules (CAMs) on umbilical cord blood (UCB) hemopoietic cells (HPCs) expanded in vitro and the optimal time for transplantation. Methods Long term culture (LTC) was used to expand the UCB HPCs. The expression of CAMs were determined by flow cytometry. Results CD11b+ cells increased significiantly at the 10th day, and quickly increased to 34.3 96 at the 30th day. CD54+ cells increased greatly at the 5thday, and reached 14.9 96 at the 30th day. CD44+ cells reached higher level at the 20th day,while CD34+ cells reached highest level at the 15th day, then decreased gradually but maintained at a higher level than that at the 5th day. Linear regression analysis showed there were interrelated between the expression of CD54 or CD44 and CD34 + HPCs. Conclusion The expression of CD54 and CD44 on UCB HPCs could play a positive role in the homing of UCB HPCs after its transplantation,the suitable time for transplantation is at the 14th day.
