摘要:

目的  探讨维生素D受体(vitamin D receptorVDR)基因多态性与代谢综合征(metabolic syndromeMetS)遗传易感性的关系。

方法 截止20213月,联合检索PubMed、中国知网、万方和中国生物医学文献数据库中有关FokI rs2228570 GABsmI rs1544410 GAMetS易感性关联的研究。采用Stata12.0统计软件进行Meta分析,以合并优势比(odds ratioOR)及其95%CI评估关联强度。

结果 最终纳入9篇文献,包括4 239例研究对象。Meta分析结果显示,FokI rs2228570多态性可显著增加MetS遗传易感性,在等位基因模型(f vs. F)、显性模型(ff+Ff vs. FF)、隐性模型(ff vs. Ff+FF)及纯合子模型(ff vs. FF)下,OR及其95%CI分别为1.251.07~1.46)、1.251.09~1.44)、1.401.10~1.77)、1.511.13~1.03);BsmI rs1544410多态性可显著降低MetS遗传易感性,在等位基因模型(b vs. B)及杂合子模型(Bb vs. BB)下,OR及其95%CI分别为0.860.76~0.98)、0.770.62~0.95)。

结论 VDR基因多态性与MetS遗传易感性关系密切。FokI f等位基因可能是MetS发病的危险因素,BsmI b等位基因可能是MetS发病的保护因素。

Abstract:

Objective To explore the relationship between the gene polymorphism of vitamin D receptor(VDR) and genetic susceptibility to metabolic syndrome(MetS).

Methods Pubmed, CNKI, Wanfang and Chinese Biomedical Literature Database were searched to obtain the related articles about the relationship between FokI rs2228570 GA, BsmI rs1544410 GA polymorphism and MetS susceptibility up to March 2021. Stata12.0 statistical software was used for meta-analysis. Combined odds ratio(OR) and its 95% confidence interval(95%CI) were used to evaluate the correlation strength.

Results A total of 9 articles with 4 239 subjects were included. The results of meta-analysis showed that the FokI rs2228570 polymorphism could significantly increase the genetic susceptibility to MetS; in allele model(f vs. F), dominant model(ff+Ff vs. FF), recessive model(ff vs. Ff+FF) and homozygous model(ff vs. FF), the OR and 95%CI were 1.25(1.07-1.46), 1.25(1.09-1.44), 1.40(1.10-1.77) and 1.51(1.13-1.03), respectively. The BsmI rs1544410 polymorphism could significantly decline the genetic susceptibility to MetS; in allele model(b vs. B) and heterozygous model(Bb vs. BB), the OR and 95%CI were 0.86(0.76-0.98) and 0.77(0.62-0.95), respectively.

Conclusion VDR gene polymorphism is significantly related to genetic susceptibility to MetS. FokI f allele may be a risk factor for MetS, and BsmI b allele may be a protective factor for MetS.
