The effects of bone morphogenetic protein9 on estradiol and progesterone production in human luteinized granulosa cells in vitro#br#

Abstract: [Abstract] Objective〖HTSS〗〓To investigate the effect of bone morphogenetic protein9(BMP9) on the secretion of estrogen(E2) and progesterone(P) in human ovarian luteinized granulosa cells.
〖HTH〗〖WTHZ〗Methods〖HTSS〗〓Collect granulosa cells in patients undergoing in vitro fertilization embryo transfer(IVFET). Human ovarian luteinized granulosa were cultured with medium plus variousc oncentration of BMP9(0, 3, 6, 12 μg/L)and folliclestimulating hormone(FSH) 75 U/L or alone for 72 h. The media were collected to measure the concentration of E2 and P by enzyme linked immunosorbent assay. And Westernblot method was used to determine the relative expression of aromatase P450(P450 arom) and steroidogenic acute regulatory protein(StAR) proteins.
〖HTH〗〖WTHZ〗Results〖HTSS〗〓Addition of different concentrations of BMP9 alone had no significant effect on E2 and P production by luteinized granulosa cells. Different concentrations of BMP9 cocultured with FSH had no significant effect on the secretion of E2, but the secretion of P was decreased with the increase of BMP9 concentration. Addition of different concentrations of BMP9 alone had no significant effect on P450 arom and StAR protein production by luteinized granulosa cells. Different concentrations of BMP9 cocultured with FSH had no significant effect on the expression level of P450 arom protein,but the expression level of StAR protein was decreased with the increase of BMP9 concentration.
〖HTH〗〖WTHZ〗Conclusion〖HTSS〗〓BMP9 had no significant effect on granulosa cells secreting E2 and P in the absence of FSH stimulation. BMP9 inhibited FSHinduced P biosynthesis and had no significant effect on E2. BMP9 may act as a luteinizing inhibitor and prevent follicles from advancing, and regulate the development of follicles.
