Abstract ] Objective Toobservetheeffectsof2-methoxyestradiol ( 2-ME ) onproliferation ,
invasion , migrationofmalignantmelanoma ( MM ) B16cells.Methods MMB16cellsculturedin
vitrowereregardedasthecontrolgroup.MMB16cellsweretreatedwithdifferentconcentrations
of2-ME ( 10 , 20 , 40 μ mol
/ L ) .MMB16cellswererecordedinMMB16-negativecellswithoutany
treatment.MorphologicalchangesofMMB16cellswereobservedbyinvertedmicroscopeafter48
h.Theplatecolonyassaywasusedtotestifythecolony-formationabilityofMMB16cellstreated
with2-methoxyestradiolfor2 weeks.Thetranswellassay wasusedtodetectthecapacityof
invasion and migration of MM B16 cells treated with 2-methoxyestradiolin different
concentrations.ThewoundhealingassaywasusedtotestifythecapacityofmigrationofMMB16
cellstreatedwith2-MEattheconcentrationsof10 μ mol
/ Lfor24h.Results Comparedwiththe
controlgroupthecolony-formationabilityofMM B16cellstreatedwith2-MEwassignificantly inhibited ( P <0.05 ) .ThecapacityofinvasionandmigrationofMM B16cellstreatedwith2-ME
wasinhibitedcomparedwiththecontrolgroup ( P <0.05 ) .Thecellsinthecontrolgroupalmost
migratedtoallthescratches.Onlysomescratchesareaswereoccupiedbycellsinthe10 μ mol
/ L
experimentalgroup.Conclusion 2-MEcouldinhibittheproliferationofMM B16cellinatime-
dose-dependentmanner.2-MEcouldinhibitthecloningabilityof MM B16cell.2-MEcould
inhibitthecapacityofinvasionandmigrationofMMB16cell.
