Effect of changes in dietary fat content on the ghrelin levels in diabetic rats

Abstract ] Objective Tostudytheregulationofdietaryfatindiabeticratsontheweight ,
differentpartsofthefatandghrelin.Methods SixtymaleSDratsweredividedintotwogroups
randomly : 15normalcontrolratsand45testrats.Thetestratswerefedwithhighfatdietfor
eightweeksandasingleintraperitonealinjectionofstreptozotocin ( STZ , 30mg /
kg
) toestablish
ananimalmodelofdiabetes.36diabeticratmodelwassuccessfullyestablished.Thediabeticrats
wererandomlydividedintofollowinggroups : 18generaldietdiabeticratsand18high-fatdiet
diabeticrats.Thenormalcontrolratsandgeneraldietdiabeticratswerefedwithgeneraldiet.The
high-fatdietdiabeticratswerefedwithhigh-fatdiet.Aftersixweeks , allratswerekilled.Blood
andadiposetissuesampleswerecollectedinallrats.Serum ghrelinwasdetectedbyenzyme-
linkedimmunoassay.Results Thegeneraldietdiabeticratsandhigh-fatdietdiabeticratswere
significantlyhigherin weight , epididymalfat , epinephrinefatand mesentericfatthannormal
controlrats , andweresignificantlylowerinplasmaghrelinconcentrationthannormalcontrol
rats.Conclusion Changesindietaryfatcontentplayaregulatoryroleonlevelsofghrelinin
plasmathataffectweightandenergyintake.
