The correlation between the expression of miR-141-3p and Cisplatin chemosensitivity of ovarian cancer cells#br#

Abstract: [Abstract] Objective To investigate the expression and role of miR-141-3p in Cisplatin resistance of ovarian cancer cells.
Methods RT-qPCR was performed to identify the expression of miR-141-3p in A2780/DDP cells. CCK8 assay was used to exam cell proliferation. Western blot analysis was used to detect proliferating cell nuclear antigen(PCNA) protein level.
Results miR-141-3p was upregulated in DDP-resistance A2780/DDP cells compared with DDP-sensitive A2780 cells(P<0.05). Overexpression of miR-141-5p in A2780 cells increased the IC50 of DDP compared with negative control, and also increased PCNA protein expression(P<0.05). Knockdown of miR-14-5p in A2780/DDP cells decreased the IC50 of DDP compared with negative control, and also decreased PCNA protein expression(P<0.05).
Conclusion The expression of miR-141-3p increased in DDP-resistance of ovarian cancer. Silencing of miR-141-3p can increase the chemo-sensitivity in ovarian cancer cells to DDP.
