Human umbilical cord mesenchymal stem cells differentiation into neuronlike cells with Baicalin in vitro

〖HTH〗目的〖HTSS〗〖KG*2〗探讨黄芩苷诱导人脐带间充质干细胞(human umbilical cord mesenchymal stem cells, hUMSCs)向神经细胞诱导分化的作用、条件和机制。
〖HTH〗方法〖HTSS〗〖KG*2〗选取培养P4 代的hUMSCs,根据黄芩苷给药情况分为4组:25 μg/mL组、50 μg/mL组、100 μg/mL组和对照组。动态观察细胞形态变化,每天进行免疫组织化学染色1次,检测诱导细胞中神经干细胞标志物巢蛋白(Nestin)、星形胶质细胞标志物胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)和神经元标志物神经元特异性烯醇化酶(neuronspecific enolase,NSE)的表达情况。
〖HTH〗结果〖HTSS〗〖KG*2〗黄芩苷诱导1 d,100 μg/mL组可见个别细胞出现了形态改变,即胞体收缩、细胞变圆、折光度增加,随时间延长逐渐形成突起,4 d呈典型神经样细胞改变并数量逐渐增多;50 μg/mL组3 d才开始出现折光度增强的类似改变,之后出现类似趋势;25 μg/mL组和对照组细胞形态未见明显变化;7 d时神经样变化的细胞可见漂浮死亡现象,神经样细胞比例下降。对照组免疫组织化学染色呈阴性。
〖HTH〗结论〖HTSS〗〖KG*2〗黄芩苷可在体外定向诱导hUMSCs向神经样细胞分化,且表达神经干细胞标志物Nestin、神经元标志物NSE以及神经胶质细胞标志物GFAP,在25~100 μg/mL浓度范围内,有随浓度增加诱导作用增强的趋势。

Abstract: [Abstract] Objective〖HTSS〗〓To investigate the effects of Baicalin on the differentiation of human umbilical cord mesenchymal stem cells(hUMSCs) into nerve cells.
〖HTH〗〖WTHZ〗Methods〖HTSS〗〓The cultured P4 generation was selected as the study subjects and divided into four groups according to the drug administration:25 μg/mL group, 50 μg/mL group, 100 μg/mL group and control group. Cell morphology was observed dynamically, and the expression of neural stem cell marker, Nestin, astrocyte marker, glial fibrillary acid protein(GFAP), and neuronal marker, neuronspecific enolase(NSE) were detected by immunohistochemistry staining daily in each group.

〖HTH〗〖WTHZ〗Conclusion〖HTSS〗〓Baicalin can induce HUMSCs differentiating into neurolike cells in vitro, and the expression of neural markers Nestin, NSE and GFAP showed an increasing tendency follow the concentration increasing in the range of 25-100 μg/mL of Baicalin.
