〖HTH〗目的〖HTSS〗〖KG*2〗探讨分泌型卷曲相关蛋白1(secreted frizzledrelated protein 1,SFRP1)及分泌型卷曲相关蛋白2(secreted frizzledrelated protein 2,SFRP2)基因启动子区甲基化状态与肾透明细胞癌(renal clear cell carcinoma,RCCC)的关系。
〖HTH〗方法〖HTSS〗〖KG*2〗采用甲基化特异性PCR(methylation specific PCR,MSP)法检测RCCC及其癌旁组织中SFRP1和SFRP2基因的甲基化情况;采用免疫组织化学SP法检测Wnt通路中心因子βcatenin蛋白表达。
〖HTH〗结果〖HTSS〗〖KG*2〗122例RCCC组织SFRP1基因和SFRP2基因甲基化发生率显著高于癌旁组织(P<005)。临床分期Ⅳ期RCCC组织SFRP1基因的甲基化发生率高于临床分期Ⅰ、Ⅱ、Ⅲ期(P<005)。RCCC组织中βcatenin蛋白的表达率明显高于癌旁组织(P<005)。高甲基化SFRP1基因在RCCC组织中和高甲基化SFRP2基因在RCCC组织中βcatenin蛋白的表达率明显高于未甲基化的RCCC组织 (P<005)。
〖HTH〗结论〖HTSS〗〖KG*2〗SFRP1、SFRP2基因甲基化可能参与由Wnt/βcatenin信号转导通路介导的RCCC的发生;SFRP1基因高甲基化可能与RCCC的侵袭及转移相关。

Abstract: [Abstract]〓Objective〖HTSS〗〓To investigate the promoter methylation status of secreted frizzledrelated protein 1 (SFRP1) and SFRP2 gene in renal clear cell carcinoma(RCCC).
〖HTH〗〖WTHZ〗Methods〖HTSS〗〓MSP was used to examine the methylation status of SFRP1 and SFRP2 gene in RCCC tissues and corresponding adjacent noncancerous tissues. The expression of βcatenin in Wnt pathway was detected by immunohistochemical streptavidin/peroxidase(SP) method.
〖HTH〗〖WTHZ〗Results〖HTSS〗〓Methylation frequencies of SFRP1 and  SFRP2 gene in 122 cases of RCCC were significantly higher than that in corresponding adjacent noncancerous tissues(P<005). The incidence of methylation of SFRP1 gene in stage Ⅳ was significantly higher than that in stage Ⅰ, Ⅱ and Ⅲ in RCCC tissues(P<005). The expression of βcatenin in RCCC tissues was significantly higher than that in corresponding adjacent noncancerous tissues(P<005). The expression of βcatenin in hypermethylated SFRP1 and SFRP2 gene in RCCC tissues was significantly higher than that in unmethylated RCCC tissues(P<005).
〖HTH〗〖WTHZ〗Conclusion〖HTSS〗〓SFRP1, SFRP2 gene methylation may be involved in the occurrence of RCCC mediated by Wnt/βcatenin signal transduction pathway, and high methylation of SFRP1 gene was associated with invasion and metastasis of RCCC.
