Abstract ] Objective Toinvestigatetheeffectofmagnesium onproliferationandnerve
growthfactor ( NGF ) secretioninSchwanncells ( SCs ) .Methods 3~5daysoldSDratswere
used.TheSCswereisolatedfromsciaticnerves invitro .Thepositivecellswerecountedbyusing
S-100immunocytochemicalstaining.SCsweredividedintofourgroups.Theculturemediumwas
DMEMcontaining10% FBSanddifferentconcentrationsofMgCl 2 ( 0 , 0.5 , 1 , 2mmol
/ L ) .MTT
wasusedtodetectthecellproliferationoftheSchwanncellsat2 , 4 , 6 , 8d.Theexpressionof
NGFwasdetectedbyELISA.Results Mostcollagenoblastwereremovedbydifferentialadhesion
about30 minutes.Theprimary-culturedSCswereadherenttotheplasticsurfaceofthecell
culturedishandgrowed24hafterinoculation.Inthe72thhourofcultivation , thecellsshowed
thetrend ofparallelarrangement.Inthe7th day , wecouldseecellaggregation undera
microscope.Throughcountingbyimmunohistochemistrytest , thepurityofSCsreached91% 6
daysaftercultivation.ThepurityofSCs was91% by S-100immunocytochemicalstaining.
Comparedwithnormaltraininggroup , 0.5mmol
/ LMgCl
2 grouphadapromotingeffectonSCs
proliferationandNGFsecretion.Thereweresignificantdifferencesin4groups , indifferenttime
points , andintheinteractionofthegroups vs timepoints (
P <0.05 ) .Conclusion Appropriate concentrationofMgCl 2 canstimulatetheproliferationandNGFsecretionofSCsofrat.
