去甲斑蝥素对人黑色素瘤A375细胞周期、活性氧自由基及细胞凋亡的影响

Effects of norcantharidin on the cell cycle, reactive oxygen species and apoptosis in human melanoma cell strain A375

  目的  探讨去甲斑蝥素(Norcantharidin,NCTD)对人黑色素瘤A375细胞周期、ROS自由基及细胞凋亡的影响及其机制。
  方法  培养人A375黑素瘤细胞,分别采用10,20,40 μmol/L NCTD预处理,培养48 h后,采用流式细胞术测定NCTD对人A375黑素瘤细胞周期的影响,采用免疫印迹法检测NCTD对人A375黑素瘤细胞蛋白Cyclin D1和P21表达的影响,采用流式细胞术测定NCTD对人A375黑素瘤细胞ROS及凋亡水平的影响。
  结果  10,20,40 μmol/L NCTD组G1期细胞比例高于对照组,S期和G2期细胞比例低于对照组,20,40 μmol/L NCTD组G1期细胞比例高于10 μmol/L NCTD组,S期和G2期细胞比例低于10 μmol/L NCTD组,40 μmol/L NCTD组G1期细胞比例高于20 μmol/L NCTD组,S期和G2期细胞比例低于20 μmol/L NCTD组(P<0.05)。40 μmol/L NCTD组处理人A375黑色素瘤细胞48 h后,细胞周期蛋白Cyclin D1、Cyclin A和CDK2低于对照组,P21蛋白高于对照组(P<0.05)。10,20,40 μmo/L NCTD组细胞内ROS水平高于对照组,20,40 μmol/L NCTD组细胞内ROS水平高于10 μmol/L NCTD组,40 μmol/L NCTD组细胞内ROS水平高于20 μmol/L NCTD组(P<0.05)。10,20,40 μmol/L NCTD组细胞凋亡率高于对照组,20,40 μmol/L NCTD组细胞凋亡率高于10 μmol/L NCTD组,40 μmol/L NCTD组细胞凋亡率高于20 μmol/L NCTD组(P<0.05)。NAC组细胞凋亡率低于对照组,NCTD组和NAC+NCTD组细胞凋亡率高于对照组,NAC+NCTD组细胞凋亡率低于NCTD组,差异均有统计学意义(P<0.05)。
  结论  NCTD通过阻滞细胞G1周期、减少S期抑制人A375黑色素瘤细胞的增殖,通过诱导ROS水平升高诱导细胞凋亡。

Abstract: [Abstract]Objective〖HTSS〗To investigate the effects of norcantharidin(NCTD) on the cell cycle, reactive oxygen species(ROS) and apoptosis in in human melanoma cell strain A375.
  〖WTHZ〗Methods〖HTSS〗The cell cycle of human A375 melanoma cells was determined by flow cytometry after 48 hours of incubation with 10, 20 and 40 μmol/L NCTD. The effects of NCTD on the expression of Cyclin D1 and P21 protein were measured by Western blot. The effects of reactive oxygen species and apoptosis on the cell cycles of A375 was measured by flow cytometric assay.
  〖WTHZ〗Results〖HTSS〗Flow cytometry showed that A375 cells treated with NCTD were blocked at G1 phase in the cell cycles in a dose dependent manner(P<0.05). The expression of Cyclin D1,Cyclin A, CDK2 protein was decreased in malignant melanoma cells treated with NCTD, however P21 protein was increased(P<0.05). The levels of ROS and apoptosis were increased in malignant melanoma cells treated with NCTD in a dose dependent manner(P<0.05). While the levels of apoptosis was decreased after treated with the ROS scavenger N-acetylcysteine(P<0.05).
  〖WTHZ〗Conclusion〖HTSS〗NCTD can inhibit the proliferation of human A375 melanoma cells by blocking the cell cycle of G1, reducing cell cycle of S and can induce the apoptosis of human A375 melanoma cells by inducing a higher level of ROS.
