卵巢癌腹水中不同肿瘤相关性巨噬细胞亚型IL10、IL12表达的差异

摘要: [摘要]目的研究不同临床分期的卵巢癌患者腹水或腹腔冲洗液中M2/M1比值;采用流式细胞学方法分选出M1型巨噬细胞和M2型巨噬细胞;检测与M1、M2相关的细胞因子白细胞介素10(interleukin10,IL10)、IL12 mRNA的相对表达量,从细胞因子水平初步阐述卵巢癌腹腔播散和转移的免疫学机制。方法①用CD64、CD163分别标记M1型巨噬细胞和M2型巨噬细胞,观察10例卵巢癌患者及17例卵巢良性肿瘤患者腹水或腹腔冲洗液中M2/M1比值。②用流式细胞学方法分选出M1型巨噬细胞和M2型巨噬细胞。③采用RTPCR方法检测检测M1型巨噬细胞、M2型巨噬细胞中IL10、IL12 mRNA的相对表达量。结果对照组M2/M1比值低于Ⅰ+Ⅱ期和Ⅲ+Ⅳ期,卵巢癌Ⅰ+Ⅱ期M2/M1比值低于Ⅲ+Ⅳ期,差异均有统计学意义(P<005)。IL10在M2型巨噬细胞中的相对表达量高于M1型巨噬细胞,IL12在M1型巨噬细胞中的相对表达量高于M2型巨噬细胞,差异均有统计学意义(P<005)。结论在上皮性卵巢癌患者腹水中有巨噬细胞浸润,早期患者M1型巨噬细胞所占比例较高,晚期患者M2型巨噬细胞所占比例较高;M2型巨噬细胞比例较高提示卵巢癌期别较高;IL10、IL12的表达差异提示M2型巨噬细胞可能有助于卵巢癌腹腔的播散及远处转移。

Abstract: [Abstract] ObjectiveTo detecte M2/M1 ratio of ascites or peritoneal washings of different macrophages subtypes of ovarian cancer patients at different stages, to separate M1 subtype macrophages and M2 subtype macrophages by flow cytometry, to test the mRNA expression of M1 and M2related cytokines IL10 and 1L12, and to explain the immune mechanism of ovarian cancer abdominal metastasis at the point of cytokines. Methods①M1 macrophages and type M2 macrophages were labeled with CD64 and CD163 respectively, and the ratio of M2/M1 in ascites or peritoneal lavage fluid in 10 cases of ovarian cancer and 17 cases of benign ovarian tumor was observed. ②Flow cytometry was used to separate M1 subtype macrophages and M2 subtype macrophages. ③RTPCR was used to test the mRNA expression of M1 and M2related cytokines IL10 and 1L12.
