Observation of the expression of VEGF, flk1, bax and bcl2 in the retina of different types of DR rat model

Abstract: [ Abstract ] Objective To observethevariation ofvascularendothelialgrowthfactor
( VEGF ), flk-1 , bax , bcl-2expressioninPDRratretinaeatablishedbysymphysialinjectionand
todefiniteitssuperiority.Methods Sprague-Dawleyratswere madeasdiabeticretinopathy
modelsbysolitaryinjection withstreptozotocinintraperitoneallyorsymphysialinjection with
streptozocinintraperitoneally and VEGFintravitreouslyrespectively.Usingfactorialdesign
empiricalmethod , weobservedthevariationofVEGF , flk-1 , bax , bcl-2expressioninretinasof
diabeticratsestablished by different methodsbyimmunohistochemistry and measuredthe
averageopticaldensityvalueofVEGF , flk-1 , bax , bcl-2inretinalganglioncelllayerandinner
plexiformlayerbyMIAS1998typeimageanalysissystem.Results VEGF , flk-1 , bax , bcl-2were
expressedineachlayerofretinainbothgroupsexceptthelayerofrodsandcones.Thefrontthree
targetswereexpressedstrongestintheganglionandnervefiberlayer , whilebcl-2wasexpressed
thestrongestininnersegmentoftherodsandconeslayer.ThenextstrongestlayerofVEGF expressionwasthe Müllercells whilethenextstrongestlayerofbcl-2expression wasthe
ganglionand nervefiberlayer.Alltargetsexpressioninsymphysialinjection groups were
strongerthanthoseinsolitaryinjectiongroups.Bcl-2expressioninganglionandnervefiberlayer
at8 weeksafter VEGFinjection wasstrongerthanthatat2 weeks.Thestrongestbcl-2
expression wasininnerplexiform layerat8 weeksinsolitaryinjection groups , whilein
symphysialinjectiongroupsitwasinganglionandnervefiberlayerat2 weeksafterVEGF
injection.Conclusion PDRrat modelestablishedbysymphysialinjectioncanrepresentthe
pathologicalchangesofhumanPDRandhasitsscientificresearchworth.
