Expression of CIAPIN1 gene in bladder cancer and its regulation on the proliferation of T24 cells#br#

Abstract: [Abstract]Objective〖HTSS〗To investigate the expression of cytokine-induced apoptosis inhibitor 1(CIAPIN1) gene in bladder cancer and adjacent tissues and its effect on the proliferation of bladder cancer T24 cells.
〖HTH〗〖WTHZ〗Methods〖HTSS〗Ninety-eight specimens of bladder cancer tissues and adjacent tissues were collected. The expression of CIAPIN1 mRNA and protein in tissue samples was detected by real-time quantitative PCR and Western blot. Knockdown of CIAPIN1 gene in bladder cancer T24 cells by siRNA.  Then MTS assay was used to detect cell proliferation.
〖HTH〗〖WTHZ〗Results〖HTSS〗The expression of CIAPIN1 mRNA in bladder cancer tissues was significantly higher than that in adjacent tissues(P<0.05). The expression of CIAPIN1 is independent of the patient's gender and age. However, the expression of CIAPIN1 was related to histological grade and TNM staging. The expression of CIAPIN1 in grade Ⅱ-Ⅲ bladder cancer patients was higher than that in grade Ⅰ. The expression of CIAPIN1 in TNM stage T2-T4 bladder cancer patients was higher than that in Ta-T1(P<0.05). Western blot analysis showed that the expression of CIAPIN1 protein in bladder cancer tissues was significantly higher than that in adjacent tissues, but no changed about that of β-actin(P<0.05). MTS results showed that knockdown of CIAPIN1 significantly inhibited the OD value of T24 cells over time compared to the control group. At 0 h, there was no difference in OD values between the control group and the knockdown group(P>0.05). The proliferation activity of CIAPIN1 gene T24 cells was significantly lower than that of the control group at 12 h, 24 h, and 48 h(P<0.05).
〖HTH〗〖WTHZ〗Conclusion〖HTSS〗CIAPIN1 may promote the progression of bladder cancer by regulating cell proliferation.
