大鼠骨组织中IRS2 mRNA水平与2型糖尿病性骨质疏松发病的关系

〖HTH〗目的〖HTSS〗〖KG*2〗研究胰岛素受体底物2(insulin receptor substrate2,IRS2)在2型糖尿病骨质疏松大鼠发病中的作用。
〖HTH〗方法〖HTSS〗〖KG*2〗64只3月龄雌性Wistar大鼠随机分为4组:正常对照(normal control,NC)组、去卵巢(normal ovariectomy group,NOVX)组、2型糖尿病(diabetes mellitus,DC)组和2型糖尿病合并去卵巢(diabetes ovariectomy group,DOVX)组,每组16只。NC组和NOVX组大鼠用常规饲料喂养,DC组和DOVX组大鼠用高脂饲料喂养(含45%高脂)。8周后,DC组和DOVX组腹腔注射链脲佐菌素。1周后,NOVX组和DOVX组切除双侧卵巢。继续喂养12周,成功建立2型糖尿病性骨质疏松大鼠模型; 分别检测各组大鼠体重、空腹血糖及胰岛素水平,计算胰岛素敏感指数;检测卵巢切除手术0周、4周、8周和12周各组大鼠骨密度;分别在卵巢切除手术4周、8周和12周时,收集股骨和肱骨组织,提取RNA,使用实时定量PCR方法检测大鼠骨组织中IRS2 mRNA水平。
〖HTH〗结果〖HTSS〗〖KG*2〗DC组和DOVX组体重均较NC组和NOVX组降低;DC组和DOVX组空腹血糖和空腹胰岛素较NC组和NOVX组增加,DC组和DOVX组胰岛素敏感指数均较NC组和NOVX组降低;去卵巢后8周和12周时,NOVX组和DOVX组骨密度和IRS2 mRNA水平均较NC组下降。
〖HTH〗结论〖HTSS〗〖KG*2〗在大鼠2型糖尿病性骨质疏发病过程中,随着骨密度降低,骨组织中IRS2 mRNA水平降低。

Abstract: [Abstract] Objective〖HTSS〗〓To investigate the role of insulin receptor substrate2(IRS2) in the pathogenesis of type 2 diabetic osteoporosis in rats.
〖HTH〗〖WTHZ〗Methods〖HTSS〗〓Sixtyfour  female Wistar rats(3months old) were randomly divided into 4 groups: normal control group(NC group), normal ovariectomy group(NOVX group), type 2 diabetes mellitus group(DC group) and diabetes ovariectomy group(DOVX group). Rats in NC and NOVX groups were fed with regular diet, while rats in DC and DOVX groups were fed with high fat diet(45%). After 8 weeks of feeding, DC and DOVX groups received an intraperitoneal injection of streptozotocin. One week later, rats in the NOVX and DOVX groups underwent ovariectomy. After 12 weeks of continuous feeding, the rat model of type 2 diabetic osteoporosis was successfully established. Body weight, fasting blood glucose, fasting insulin and insulin sensitivity index were measured. Bone mineral density and IRS2 mRNA levels were measured at 4, 8 and 12 weeks after ovariectomy.
〖HTH〗〖WTHZ〗Results〖HTSS〗〓Compared with NC group and NOVX group, the body weight of DC group and DOVX group was decreased, while the fasting blood glucose and insulin were increased. Compared with NC group and NOVX group, insulin sensitivity index of DC group and DOVX group was decreased. Compared with NC group, the bone mineral density and IRS2 gene levels of NOVX group and DOVX group were gradually decreased at 8 and 12 weeks after ovariectomy.
〖HTH〗〖WTHZ〗Conclusion〖HTSS〗〓IRS2 mRNA level in bone tissue decreases with the decrease of bone density during the development of type 2 diabetic osteoporosis.
