大鼠心肌梗死后血清脑钠肽、内皮素1和一氧化氮的变化及相关性研究

摘要: 目的:观察急性心肌梗死(acute myocardial infarction,AMI)后大鼠脑钠肽( brain natriuretic peptide, BNP)、内皮素1(endothelin-1,ET-1)和一氧化氮(nitrogen monoxide,NO)的变化以及相关性。方法 Wistar 大鼠80只,经麻醉后行冠状动脉结扎处理制作心肌梗死模型,术后存活的大鼠分为对照组( Sham,n =40)和心肌梗死组(AMI,n =40)。术后开始计时,分别于6h、12h、24h、3d、7d 和30d 抽取大鼠静脉血1mL,测定 BNP、ET-1和 NO 水平,并分析三者的相关性。结果2组 BNP、ET-1和 NO 水平在组间、不同时点以及组间和不同时点的交互作用差异均有统计学意义(P ﹤0.01);相关性分析提示 ET-1与 NO 呈负相关(r =-0.948,P =0.004)。结论 AMI 后出现了血清 BNP、ET-1升高和 NO 降低的变化。为 AMI 后神经内分泌激素的变化提供了理论依据。

Abstract: Objective To explore serum levels of brain natriuretic peptide(BNP),endothelin-1 (ET-1)and nitrogen monoxide(NO)in rats after acute myocardial infarction(AMI)and to observe their correlation. Methods Eighty male Wister rats were randomly divided into two groups:sham group(n =40),AMI group(n = 40). The serum concentrations of BNP,ET-1 and NO were measured at different time(6h,12h,24h,3d,7d and 30d)to compare the difference between two groups,and their correlation were analyzed. Results There were significant difference in the serum levels of BNP,ET-1 and NO between AMI group and sham group(P ﹤ 0. 01). For different groups,there were significant differences in the serum levels of BNP,ET-1 and NO among different time(P ﹤ 0. 01). From the interactional effort of group and time,there were different evolvement in the serum levels of BNP,ET-1 and NO between two groups(P ﹤ 0. 01). The analysis in AMI group showed that serum ET-1 was negatively correlated with serum NO. Conclusion This study showed serum BNP and ET-1 increase and serum NO decrease after AMI,which may lay theory evidence for neural endocrine hormone change.
