Expression of SNAI2 in human malignant glioma and its relationship with cell proliferation and metastasis#br#

Abstract: [Abstract] Objective〖HTSS〗〓To investigate the mRNA expression level of zincfinger transcription factor2(SNAI2) gene in clinical tissues, and to identify the effect of SNAI2 on the proliferation and invasion of glioma cancer cells U251.
〖HTH〗〖WTHZ〗Methods〖HTSS〗〓A total of 37 cases of patients with malignant glioma was selected in our hospital. Reverse transcription polymerase chain reaction(RTPCR) was used to detect the mRNA level of SNAI2 in tissues and to analyze the relationship between the expression and the clinical pathological data.In vitro, we set up a stable SNAI2 interference(shSNAI2) of glioma cells. The ability of cell proliferation was detected by MTT colorimetric assay(MTS). The ability of cell invasion and metastasis was detected by scratch assay, Western blot was used to detect the expression of Ecadherin, vimentin and Zo1, related with epithelial mesenchymal transition(EMT).
〖HTH〗〖WTHZ〗Results〖HTSS〗〓The expression of SNAI2 in highgrade gliomas was significantly higher than that in low grade gliomas, benign meningiomas and normal brain tissue, and the expression of SNAI2 in low grade gliomas was significantly higher than that in benign meningiomas and normal brain tissue, and the difference was statistically significant(P<005). Spearman correlation analysis was used to compare the relationship between the expression of SNAI2 and the clinical pathological data of patients, and the results show that the expression of SNAI2 and the patient's gender, age and body weight were not statistically related. There was a positive correlation between tumor size and tumor size and tumor grade(P<005). The results of MTS experiment showed that the proliferation ability of cells in each group increased with time, and the proliferation ability of shSNAI2 cell line was significantly lower than that of control cell line(P<005). The results of scratch repair showed that the ability of cell migration of shSNAI2 cell line was significantly less than that of control cell line. The results of Western blot showed that the expression of Ecadherin and Zo1 in shSNAI2 cell line was increased, and the expression of Vimentin was decreased(P<005), and the difference was statistically significant(P<005).
〖HTH〗〖WTHZ〗Conclusion〖HTSS〗〓SNAI2 is highly expressed in human brain gliomas, and is closely related to the size and grade of tumor. It can promote the proliferation and metastasis of glioma cells and the occurrence of EMT.
