〖HTH〗目的〖HTSS〗〖KG*2〗探讨电刺激对大鼠坐骨神经卡压损伤后神经再生和恢复的作用。
〖HTH〗方法〖HTSS〗〖KG*2〗选用健康雄性大鼠,制备经典大鼠坐骨神经慢性卡压模型,3个月后将损伤的坐骨神经外膜切开,置入刺激电极并皮下埋入刺激器,进行电刺激治疗12周,评估电刺激治疗对大鼠坐骨神经再生和功能恢复情况,以及患肢三头肌萎缩情况;采用神经组织染色法,用光镜和透射电镜,观察坐骨神经髓鞘的再生情况。
〖HTH〗结果〖HTSS〗〖KG*2〗周围神经慢性卡压模型建立后,大鼠坐骨神经明显受损。2组造模成功时坐骨神经功能指数(sciatic function index,SFI)指数明显降低,随电刺激治疗延长,SFI明显升高,治疗后实验组各时点SFI均高于对照组,差异有统计学意义(P<005)。造模成功时,2组大鼠肌肉均发生萎缩,重量明显降低;治疗后实验组各时点肌肉重量均大于对照组,差异有统计学意义(P<005)。随治疗时间延长,2组神经传导速度明显升高,治疗4,8,12周时实验组大鼠神经传导速度均高于对照组,差异有统计学意义(P<005)。治疗12周时2组髓神经纤维数量、神经纤维直径、髓鞘厚度均大于造模成功时,且实验组髓神经纤维数量、神经纤维直径、髓鞘厚度均大于对照组,差异有统计学意义(P<005)。
〖HTH〗结论〖HTSS〗〖KG*2〗低频电刺激治疗可促进慢性卡压损伤坐骨神经的再生和功能恢复。

Abstract: [Abstract]〓Objective〖HTSS〗〓To investigate the effect of electrical stimulation(ES) on sciatic nerve regeneration and function recovery after a crush injury in rats.
〖HTH〗〖WTHZ〗Methods〖HTSS〗〓A chronic nerve compression model was created by surgery on sciatic  nerves of male rats. After 3 months,epineurial of the injured nerves were incised, and then a flexible wireless stimulator was implanted in rats to provide electrical stimulation for 12 weeks. Thereafter, recovery of the injured sciatic nerve was assessed by the sciatic functional index(SFI), affected muscle weight. and conduction velocity of injured sciatic nerve measured by electrophysiology. Regeneration and remyelination of injured nerve were assessed by histology investigation and transmission electron microscope observation.
〖HTH〗〖WTHZ〗Results〖HTSS〗〓The sciatic nerves were clearly injured in both groups of rats, with significantly decreased SFI. During the ES treatment course, the SFI was increased significantly, and the SFI of rats with ES treatment were superior to the controls at each time points(P<005). Muscular atrophy was observed in all the involved rats with decreased weight of affected muscles, whereas the muscle weight was significantly higher in the rats received ES treatment compared to the controls(P<005). During the treatment course, nerve conduction velocity was increased in both groups of rats, which were significantly higher in the rats with ES treatment compared to the controls at the 4 th, 8 th, and 12 th week(P<005). In addition, after 12 weeks period of treatment, the ES increased nerve diameter, quantity of myelinated nerve fibers and thickness of myelin sheath(P<005).
〖HTH〗〖WTHZ〗Conclusion〖HTSS〗〓Electrical stimulation could produce a therapeutic effect on the regeneration and function recovery of injured sciatic nerves by compression.
