Effect of resveratrol on proliferation of lung cancer A549 cells and the expression of NFκB

摘要: [摘要] 目的 观察白黎芦醇对人肺癌细胞 A549 增殖及核转录因子 κB ( nuclearfactor-kappaB , NF-κB )表达
的影响。方法 选用噻唑蓝( methylthiazolyldiphenyl-tetrazoliumbromide , MTT )法检测白藜芦醇对 A549 细胞增殖
的影响;采用逆转录 PCR ( reversetranscription-PCR , RT-PCR )检测白藜芦醇对 A549 细胞 NF-κB 信使核糖核酸
( messengerRNA , mRNA )表达的作用;采用免疫印迹法检测白藜芦醇对肺癌 A549 细胞 NF-κB 蛋白水平的影响。
结果 随白藜芦醇浓度的增加, B 、 C 、 D 组细胞生长抑制率均高于 A 组,
C 、 D 组高于 B 组, D 组高于 C 组( P <0.05 )。
B 、 C 、 D 组 24 、 48h 细胞生长抑制率高于 12h , 48h 细胞生长抑制率高于 24h ( P <0. 05 )。经 100 μ mol / L 白藜芦醇
作用 48h 后, A549 细胞可以明显下调 NF-κBmRNA 和蛋白水平的表达。结论 白藜芦醇可以明显抑制肺癌 A549
细胞的增殖及 NF-κB 的表达。

Abstract ] Objective Tostudytheeffectofresveratrolonproliferationofhumanlung
cancercelllineA549andtheexpressionofnuclearfactorkappaB ( NF-κB ) .Methods Theeffect
ofresveratrolonthe proliferation of A549 cells was detected by methylthiazolyldiphenyl-
tetrazoliumbromide ( MTT ) method.TheeffectofresveratrolontheexpressionofmRNANF-κB
inA549cellswasdetectedbyreversetranscription-PCR ( RT-PCR ) .EffectofresveratrolonNF-
κBproteinlevelsinlungcancerA549cellswasdetectedbywesternblotting.Results Withthe
increaseoftheconcentrationofresveratrol , thecellgrowthinhibitionratesingroupB , CandD
werehigherthaninthatingroupA.ThecellgrowthinhibitionratesingroupCandD were
higherthanthatingroupB.ThecellgrowthinhibitionratesingroupDwashigherthanthatin
groupC ( P <0.05 ) .Cellgrowthinhibitionrateat24hand48ingroupB , CandDarehigher
thanthatat12h.Cellgrowthinhibitionrateat48hwashigherthanthatat24h ( P <0.05 ) .

NF-kappamRNA.Conclusion Resveratrolcansignificantlyinhibittheproliferationoflung cancerA549cellsandtheexpressionofNF-κB.
