姜黄素对糖尿病大鼠子宫内膜整合素β3、 LIF表达水平的影响

〖HTH〗目的〖HTSS〗〖KG*2〗观察姜黄素在治疗糖尿病及其并发症过程中对整合素β3、白细胞抑制因子(leukemia inhibitory factor,LIF)表达水平的影响。
〖HTH〗方法〖HTSS〗〖KG*2〗将雌性SD大鼠30只随机分为 3 组进行灌胃治疗,分别为:正常组10只, 选用食用玉米油灌胃10周2.5 mL/kg;糖尿病模型组10只,选用食用玉米油灌胃10周2.5 mL/kg;姜黄素治疗组 10只,选用姜黄素化合物混悬液灌胃10周每日250 mg/kg。10周后,按雌雄比例3〖DK〗∶2合笼,次日晨查雌鼠阴道,发现阴栓者计为妊娠第 1 天,雌性大鼠均于妊娠第 4 天取材。分别检测血糖、胰岛素,采用 HE 染色观察子宫内膜上皮的形态结构,运用免疫组织化学和免疫印迹实验法分析各组大鼠子宫内膜整合素β3、 LIF表达水平。
〖HTH〗结果〖HTSS〗〖KG*2〗糖尿病组与正常组比较,其血糖和胰岛素明显升高,子宫内膜整合素β3和LIF表达量明显减少,姜黄素治疗组与糖尿病组比较,其血糖和胰岛素明显降低,子宫内膜整合素β3和LIF表达量明显升高。
〖HTH〗结论〖HTSS〗〖KG*2〗姜黄素可降低血糖和胰岛素,增加子宫内膜整合素β3和LIF蛋白的表达,最终改善子宫内膜容受性。

Abstract: [Abstract] Objective〖HTSS〗〓To investigate the effect of curcumin on the expression of integrin β3 and leukemia inhibitory factor(LIF) in endometrium of diabetic rats.
〖HTH〗〖WTHZ〗Methods〖HTSS〗〓Thirty female SpragueDawley rats were divided into  control group,Diabetes model group and curcumin compound group randomly. The normal 〖JP2〗controls and diabetes model group served with edible corn oil 2.5 mL/kg for 10 weeks, while the curcumin compound group were treated with 250 mg·〖JP〗kg-1·d-1 curcumin compound suspension for the same time. After 10 weeks, the ratio of male to female was 3〖DK〗∶2 for mating. The vagina of the female was checked next day. The fetus was found to be the first day of pregnancy and were sacrificed in the fourth day. Blood glucose and insulin were measured separately. The morphology of endometrial epithelium were observed by HE staining and immunohistochemistry. Western blot analysis were used to analyze the changing of the endometrial integrin β3 and LIF after treatment with curcumin for 10 weeks.
〖HTH〗〖WTHZ〗Results〖HTSS〗〓Compared with the normal group, the blood glucose and insulin were significantly increased in the diabetic group(P<005), and the expression of integrin β3 and LIF in the endometrium were significantly decreased(P<005). Compared with the diabetes group, the blood glucose and insulin were significantly lower(P<005), and the expression of integrin β3 and LIF in the endometrium were significantly increased(P<005) in the curcumin compound treatment group.
〖HTH〗〖WTHZ〗Conclusion〖HTSS〗〓Curcumin can lower blood sugar and insulin, increase the expression of endometrial integrin β3 and LIF protein, and finally improve endometrial receptivity.
