Abstract: [
Abstract ] Objective Tostudytheinhibitioneffectsof2-methoxyestradiol ( 2-ME ) on
malignantmelanoma ( MM ) cellsinBALB / c micexenografttumor.Methods Theinhibition
effects of 2-ME on MM cells were measured by 3- ( 4 , 5-Dimethylthiazol-2-yl ) -2 , 5-
diphenyltetrazoliumbromide ( MTT ) .BuildingBALB / cmicexenografttumormodel , BALB / c
miceweredividedintothreegroupswithsixmiceineachgroup.Treatmentgroupandnegative
controlgroupweretreatedwith2-ME ( 50mg /
kg
) andequallyvolumenormalsalinerespectively
everytwodays , fora14-daysdurationandcalculatedtheaveragevolumeeverythreedays. PositivedruggroupweretreatedwithCTX ( 20 mg /
kg
) .Afterexperimentended , thetumor
weightwerecalculated.TheexpressionlevelsofC-mycandGadd45bintumorweredetectedby
immunohistochemical.Morphologicalchangesoftumortissue , lung , liverandspleentissues
wereobservedbyHEstaining.Results 2-MEhadobviousinhibitoryeffectonmelanomacells
andtheinhibitoryeffectinadose-timedependentmanner.ComparedwiththeNaclgroup , the
volumeandweightoftumortissuesinCTXgroupand2-MEgroupwerereducedremarkably
(
P <0. 01 ), buttherewasnostatisticalsignifieancebetweentheCTXgroupandthe2-MEgroup
(
P >0.05 ) .Comparingtocontrolgroup , theexpressionlevelsofC-mycweredecreased , while
Gadd45bwereincreased.Noobviousmorphologicalchangeswerefoundinliverandspleentissues
intreatmentgroupandincontrolgroup , buttherewerelungmetastasisin3of6mice.Conclusion
2-MEhadinhibitoryeffectonBALB / cmicexenografttumor , andthe mechanism maybe
relatedtomodulatinggadd45bandC-mycgenesexpression.
