Abstract: [Abstract] Objective〖HTSS〗〓To establish an effective and simple method for isolation and expansion of the adipose tissuederived stem cells(ASCs), by using the adipose tissues of abdominal cavity of Sprague Dawley rat, and to provide another choice for cell source of mesenchymal stem cells(MSCs).
〖HTH〗〖WTHZ〗Methods〖HTSS〗〓The adipose tissues were obtained from the abdominal cavity of Sprague Dawley rats aged 4 weeks. 0.1% collagenase Ⅰ was chosen for digesting the adipose tissues. After washing and centrifugation, the cells were resuspended in mesenchymal stem cell medium and cultured in cell incubator. The growth rate was assessed by MTT assay. ASCs were then verified by the flow cytometry analysis and differentiation assays.
〖HTH〗〖WTHZ〗Results〖HTSS〗〓ASCs were spindlelike in shape and adhered to the plastic tissue culture medium, with high multiplication rate. The absorbance value of passage 5 was lower passage 3, and the absorbance value of passage 8 was lower than passage 3 and passage 5 on day 2, 4, 5 and 6(P<005). ASCs were positive for CD29, CD54 and CD90.1, but negative for CD31, CD45 and CD106.
〖HTH〗〖WTHZ〗Conclusion〖HTSS〗〓ASCs can be effectively isolated and expanded from the adipose tissues of abdominal cavity of Sprague Dawley rat, by using adherent culture. And ASCs from adipose tissues of abdominal cavity may be a good multipotential cell candidate for future cell replacement therapy.
